چکیده

Our previous studies of the mechanism of cell growth inhibition by gallium have suggested that the block in cellular iron uptake induced by transferrin-gallium results in an inhibition of the iron-dependent M2 subunit of ribonucleotide reducÃ-ase.However, it is not known whether the inhibitory effect of gallium on ribonucleotide reducÃ-aseis solely the result of limiting iron availability for enzyme activity or whether a direct effect of intracellular gallium on the enzyme is also involved. In the present study, utilizing a cell-free assay, we show that gallium nitrate directly inhibits CDP and ADP reducÃ-aseactivity. Inhibition of DNA synthesis by gallium nitrate thus appears to be due to a combination of a block in iron availability to ribonucleotide reducÃ-aseand a direct inhibi tion of the enzyme by gallium.

Modulation of Transfer RNA-methylating Enzyme Activities in Murine Leukemic Cells1

The activities of those processing enzymes responsible for the methylation of tRNA in murine leukemic cells were compared in sublines selected for their sensitivity and resistance to various antineoplastic agents. With one exception, the capacity and extent of tRNA methylation were elevated in the drug-resistant sublines. The following order of activities was demonstrated: L1210/cytoxan > L1210...

2,6-Diaminopurinedeoxyriboside as a Prodrug of Deoxyguanosine in LI 210 Cells1

The mode of action of the antiproliferative nucleoside analogue 2,6diaminopurinedeoxyriboside (DAPdR) has been characterized in cultured 11210 cells. A marked concentration-dependent decrease in DNA syn thesis and ribonucleotide reductase activity occurred in LI 210 cells exposed to 0.05 to 1.0 HIM DAPdR. Concomitantly, dGTP levels in creased as much as 1100-fold as compared to untreated contro...

Comparative Biochemical Studies of Adriamycin and Daunomycin in Leukemic Cells1

Adriamycin (AM) and daunomycin (DM) were compared as inhibitors of nucleic acid and protein biosynthesis in LI 210 leukemic cells. In vivo, both antibiotics markedly inhibited, without significant difference, the incorporation of the labeled precursors into DNA and RNA. In vitro, at a concentration of 5 or lOjUg/ml, DM was significantly more effective than AM in inhibiting DNA and RNA synthesis...

Chelators: Effects on the Proliferation of Human Leukemic Interaction of Gallium Nitrate with Fludarabine and Iron

Earlier studies have shown that transferrin-gallium inhibits cellular iron incorporation and blocks DNA synthesis by decreasing the activity of the iron-dependent M2 subunit of ribonucleotide reducÃ-ase.We ex amined the growth-inhibitory effects of gallium nitrate in combination with clinically relevant inhibitors of ribonucleotide reducÃ-asefludarabine (an Ml subunit inhibitor), and iron chela...

Inhibition of leukemic HL60 cell growth by transferrin-gallium: effects on ribonucleotide reductase and demonstration of drug synergy with hydroxyurea.

Cellular requirements for iron during DNA synthesis are related to the increased activity of the iron-containing M2 subunit of ribonucleotide reductase, the enzyme responsible for the reduction of ribonucleotides to deoxyribonucleotides. We have previously shown that transferrin-gallium (Tf-Ga) inhibits cellular iron incorporation. In the present study, Tf-Ga-induced inhibition of HL60 cell gro...
